The best guess I've seen (and at least partially verified by Mladek) is that it is a bass kazoo. My guess is that the brass spittoon thing is just there as a resonator, and since it's jug band music would occupy the same role as a stovepipe.

However, to my ears, which are still functioning, it sounds like the "burlesque" preset on a goddamn ondioline. So maybe the whole thing is a lip-synched sham.
